Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_017950.4(CCDC40):c.1949A>G (p.Asn650Ser), citing Ambry Variant Classification Scheme 2023: The p.N650S variant (also known as c.1949A>G), located in coding exon 12 of the CCDC40 gene, results from an A to G substitution at nucleotide position 1949. The asparagine at codon 650 is replaced by serine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
