Uncertain significance — the classification assigned by Ambry Genetics to Single allele, citing Ambry Variant Classification Scheme 2023: The c.63A>C (p.E21D) alteration is located in exon 1 (coding exon 1) of the LRRC7 gene. This alteration results from a A to C substitution at nucleotide position 63, causing the glutamic acid (E) at amino acid position 21 to be replaced by an aspartic acid (D). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
