This variant lies in the MYH7 gene (transcript NM_000257.4) at coding-DNA position 497, where T is replaced by G; at the protein level this means replaces leucine at residue 166 with arginine — a missense variant. Submitter rationale: This sequence change replaces leucine with arginine at codon 166 of the MYH7 protein (p.Leu166Arg). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and arginine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a MYH7-related disease. A computational algorithm designed to assess the pathogenicity of variants in MYH7 with regard to hypertrophic cardiomyopathy predicted this sequence change to be deleterious.
