Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4474C>T (p.His1492Tyr), citing Ambry Variant Classification Scheme 2023: The p.H1446Y variant (also known as c.4336C>T), located in coding exon 39 of the KIF1B gene, results from a C to T substitution at nucleotide position 4336. The histidine at codon 1446 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
