Uncertain significance — the classification assigned by Ambry Genetics to NM_006393.3(NEBL):c.2401G>A (p.Val801Met), citing Ambry Variant Classification Scheme 2023: The p.V801M variant (also known as c.2401G>A), located in coding exon 24 of the NEBL gene, results from a G to A substitution at nucleotide position 2401. The valine at codon 801 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
