NM_000245.4(MET):c.3204T>A (p.His1068Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1086Q variant (also known as c.3258T>A), located in coding exon 14 of the MET gene, results from a T to A substitution at nucleotide position 3258. The histidine at codon 1086 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
