Uncertain significance for Amyotrophic lateral sclerosis type 21 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018834.6(MATR3):c.1487G>A (p.Arg496His), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 496 of the MATR3 protein (p.Arg496His). This variant is present in population databases (rs368389424, gnomAD 0.008%). This variant has not been reported in the literature in individuals affected with MATR3-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt MATR3 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
