NM_000057.4(BLM):c.2453G>A (p.Arg818His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R818H variant (also known as c.2453G>A), located in coding exon 11 of the BLM gene, results from a G to A substitution at nucleotide position 2453. The arginine at codon 818 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
