This variant has been identified in 0.01% (5/41448) of African/African American chromosomes by gnomAD (http://gnomad.broadinstitute.org, v4.1.0) which is low enough allele frequency to be consistent with a recessive disorder. Furthermore, although this gene has been reported in association with a dominant neurodevelopmental disorder, this association has not been definitively established with a recessive mode of inheritance. In summary, the clinical significance of this variant is uncertain.
