This variant lies in the RYR1 gene (transcript NM_000540.3) at the canonical splice acceptor site of the intron immediately before coding-DNA position 10019, duplicating one base. Submitter rationale: Variant summary: RYR1 c.10019-2dupA alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 3' acceptor site and one predicts the variant weakens a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3.2e-05 in 250450 control chromosomes.
