Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 1.2e-05 in 251484 control chromosomes. c.5435T>A has been observed in individuals affected with Retinitis Pigmentosa/ABCA4-related retinopathy (e.g. Sung_2020, Wang_2022). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function and found the variant resulted in <10% of normal protein expression in vitro, suggesting it may result in an unstable protein (Xu_2023).
