This variant lies in the PPP2R5C gene (transcript NM_001352913.2) at 32903 bases into the intron immediately after coding-DNA position 259, where G is replaced by A. Submitter rationale: Variant summary: PPP2R5C c.187+1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing, however current evidence is not sufficient to establish loss of function as a mechanism for disease. Several computational tools predict a significant impact on normal splicing: three predict the variant abolishes a 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 6.5e-06 in 154060 control chromosomes (gnomAD).
