Likely pathogenic for Familial intrahepatic cholestasis; Low phospholipid associated cholelithiasis — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000443.4(ABCB4):c.1633C>T (p.Arg545Cys), citing Genomenon Sequence Variant Interpretation Standards - Updated: ABCB4 p.Arg545Cys (c.1633C>T) is a missense variant that changes the amino acid at residue 545 from Arginine to Cysteine. This variant has been observed in at least one proband with an ABCB4-related disorder (PMID:39333837;36277956;26324191). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:28220208). It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify ABCB4 p.Arg545Cys (c.1633C>T) as a likely pathogenic variant.
