Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000550.3(TYRP1):c.340C>T (p.Arg114Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TYRP1 c.340C>T (p.Arg114Cys) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 1.2e-05 in 250222 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.340C>T has been observed in at least one individual affected with Oculocutaneous albinism (Lasseaux_2018). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 29345414). No submitters have cited clinical-significance assessments for this variant to ClinVar. Based on the evidence outlined above, the variant was classified as uncertain significance.
