Likely pathogenic for Nonsyndromic genetic hearing loss — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_004004.6(GJB2):c.550C>G (p.Arg184Gly), citing LabCorp Variant Classification Summary - May 2015: Variant summary: GJB2 c.550C>G (p.Arg184Gly) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251266 control chromosomes (gnomAD). c.550C>G has been observed in an individual affected with Non-Syndromic Hearing Loss (Zoll_2003). Other variants affecting the same codon have been classified as pathogenic by our lab (c.550C>T/p.Arg184Trp, c.551G>A/p.Arg184Gln, c.551G>C/p.Arg184Pro), supporting the critical relevance of codon 184 to GJB2 protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
