Loss-of-function variants in RYR1 are known to be pathogenic (PMID: 23919265, 25960145, 28818389, 30611313). The variant was absent in control chromosomes (gnomAD). To our knowledge, no occurrence of c.12928_12988del61 in individuals affected with RYR1-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
