This variant lies in the NPC1 gene (transcript NM_000271.5) at coding-DNA position 1892, where T is replaced by G; at the protein level this means replaces methionine at residue 631 with arginine — a missense variant. Submitter rationale: Variant summary: NPC1 c.1892T>G (p.Met631Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251430 control chromosomes. c.1892T>G has been observed in compound heterozygous or heterozygous individuals affected with Niemann-Pick Disease Type C (e.g., Marelli_2016, Millat_2005, Sedel_2016, SekerYilmaz_2020). These data indicate that the variant may be associated with disease. One publication reports experimental evidence evaluating an impact on protein function, showing trafficking alterations in vitro (e.g. Shammas_2019), however, does not allow convincing conclusions about the variant effect.
