Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000051.4(ATM):c.1495C>T (p.Gln499Ter), citing ACMG Guidelines, 2015: This variant changes 1 nucleotide in exon 10 of the ATM gene, creating a premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, this variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been observed on the opposite chromosome (in trans) from a pathogenic variant in an individual with clinical features of autosomal recessive ataxia-telangiectasia (ClinVar SCV000622265.5), indicating that this variant contributes to disease. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of ATM function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic.
