Single allele was classified as pathogenic by Quest Diagnostics Nichols Institute San Juan Capistrano, citing Quest Diagnostics criteria: A deletion of a portion of exon 56 (c.8218_8268) in the ATM gene is predicted to disrupt the translation reading frame of the ATM mRNA and result in the premature termination of ATM protein synthesis. In the published literature, a similar overlapping deletion of a portion of exon 56 in the ATM gene has been reported in an individual with ataxia-telangiectasia (PMID: 26896183 (2016)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic.
