Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000136.3(FANCC):c.1370C>G (p.Ser457Cys), citing Ambry Variant Classification Scheme 2023: The p.S457C variant (also known as c.1370C>G), located in coding exon 13 of the FANCC gene, results from a C to G substitution at nucleotide position 1370. The serine at codon 457 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
