This variant lies in the GIGYF2 gene (transcript NM_001103146.3) at coding-DNA position 1283, where A is replaced by G; at the protein level this means replaces glutamic acid at residue 428 with glycine — a missense variant. Submitter rationale: The E428G variant in the GIGYF2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E428G variant is observed in 1/111562 (0.009%) alleles from individuals of European (non-Finnish) background, in large population cohorts (Lek et al., 2016). The E428G variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals.
