NM_001148.6(ANK2):c.10841A>G (p.His3614Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H3614R variant (also known as c.10841A>G), located in coding exon 40 of the ANK2 gene, results from an A to G substitution at nucleotide position 10841. The histidine at codon 3614 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
