Likely pathogenic for Neuronal ceroid lipofuscinosis 5 — the classification assigned by Kasturba Medical College, Manipal, Kasturba Medical College, Manipal, Manipal Academy of Higher Education, Manipal, India to NM_006493.4(CLN5):c.570del (p.Asn190fs), citing ACMG Guidelines, 2015: A novel frameshift deletion, c.570del in exon 4 of CLN5 was observed in homozygous state in proband. Sanger validation and segregation analysis revealed that the variant was present in homozygous state in the proband and in a heterozygous state in his parents. This variant is not reported in homozygous and/or heterozygous state in the population database gnomAD (v4.1.0) and our in-house exome database of 3840 individuals. This deletion is likely to cause a shift in the reading frame and result in a premature truncation of the transcript, which can either lead to nonsense-mediated mRNA decay or the formation of a truncated CLN5 protein product.
