This variant lies in the ACTN2 gene (transcript NM_001103.4) at coding-DNA position 244, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 82 with lysine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the ACTN2 gene. The E82K variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The E82K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. However, to our knowledge no studies have been performed to determine the functional effect of the E82K variant.
