Uncertain significance — the classification assigned by GeneDx to NM_001001344.3(ATP2B3):c.2269C>T (p.Pro757Ser), citing GeneDx Variant Classification (06012015). This variant lies in the ATP2B3 gene (transcript NM_001001344.3) at coding-DNA position 2269, where C is replaced by T; at the protein level this means replaces proline at residue 757 with serine — a missense variant. Submitter rationale: The P757S variant in the ATP2B3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The P757S variant is not observed in large population cohorts (Lek et al., 2016). The P757S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret P757S as a variant of uncertain significance.
