This variant lies in the GBE1 gene (transcript NM_000158.4) at coding-DNA position 1345, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 449 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E449X variant in the GBE1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E449X variant is not observed in large population cohorts (Lek et al., 2016). We interpret E449X as a pathogenic variant.
