This variant lies in the PDGFRA gene (transcript NM_006206.6) at coding-DNA position 701, where C is replaced by T; at the protein level this means replaces threonine at residue 234 with isoleucine — a missense variant. Submitter rationale: The T234I variant in the PDGFRA gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The T234I variant is not observed in large population cohorts (Lek et al., 2016). The T234I variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Threonine are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret T234I as a variant of uncertain significance.
