Uncertain significance — the classification assigned by GeneDx to NM_017617.5(NOTCH1):c.2443T>A (p.Cys815Ser), citing GeneDx Variant Classification (06012015): The C815S variant in the NOTCH1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The C815S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In addition, this substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret C815S as a variant of uncertain significance.
