NM_015631.6(TCTN3):c.959T>C (p.Val320Ala) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the TCTN3 gene (transcript NM_015631.6) at coding-DNA position 959, where T is replaced by C; at the protein level this means replaces valine at residue 320 with alanine — a missense variant. Submitter rationale: The V320A variant in the TCTN3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The V320A variant is observed in 46/22820 (0.2%) alleles from individuals of South Asian background, in the ExAC dataset (Lek et al., 2016). The V320A variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to Valine are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret V320A as a variant of uncertain significance.
