Likely pathogenic — the classification assigned by GeneDx to NM_001376.5(DYNC1H1):c.7064C>T (p.Thr2355Ile), citing GeneDx Variant Classification (06012015): The T2355I variant in the DYNC1H1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The T2355I variant is not observed in large population cohorts (Lek et al., 2016). The T2355I variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species, and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret T2355I as a likely pathogenic variant.
