This variant lies in the LIPC gene (transcript NM_000236.3) at coding-DNA position 759, duplicating one base; at the protein level this means shifts the reading frame starting at cysteine residue 254, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.759dupC variant in the LIPC gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.759dupC variant causes a frameshift starting with codon Cysteine 254, changes this amino acid to a Leucine residue, and creates a premature Stop codon at position 51 of the new reading frame, denoted p.Cys254LeufsX51. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay, however, loss-of-function is not a know mechanism of disease for LIPC. The c.759dupC variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.759dupC as a variant of uncertain significance.
