Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000528.4(MAN2B1):c.1502G>C (p.Cys501Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: MAN2B1 c.1502G>C (p.Cys501Ser) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 250324 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1502G>C has been observed in an individuals affected with Alpha-Mannosidosis with another VUS in cis and without a variant in trans specified (Kuokkanen_2011). This report does not provide unequivocal conclusions about association of the variant with Alpha-Mannosidosis. This publication also reports experimental evidence evaluating an impact on protein function, however, does not allow convincing conclusions about the variant effect.
