Likely pathogenic — the classification assigned by GeneDx to NM_006009.4(TUBA1A):c.916G>T (p.Asp306Tyr), citing GeneDx Variant Classification (06012015). This variant lies in the TUBA1A gene (transcript NM_006009.4) at coding-DNA position 916, where G is replaced by T; at the protein level this means replaces aspartic acid at residue 306 with tyrosine — a missense variant. Submitter rationale: The D306Y variant in the TUBA1A gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The D306Y variant is not observed in large population cohorts (Lek et al., 2016). The D306Y variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret D306Y as a likely pathogenic variant.
