NM_001999.4(FBN2):c.791G>A (p.Gly264Asp) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the FBN2 gene (transcript NM_001999.4) at coding-DNA position 791, where G is replaced by A; at the protein level this means replaces glycine at residue 264 with aspartic acid — a missense variant. Submitter rationale: The G264D variant in the FBN2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G264D variant is not observed in large population cohorts (Lek et al., 2016). The G264D variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret G264D as a variant of uncertain significance.
