This variant lies in the TBX5 gene (transcript NM_181486.4) at coding-DNA position 164 through coding-DNA position 165, deleting 2 bases; at the protein level this means shifts the reading frame starting at lysine residue 55, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.164_165delAA pathogenic variant in the TBX5 gene causes a frameshift starting with codon Lysine 55, changes this amino acid to a Serine residue and creates a premature Stop codon at position 5 of the new reading frame, denoted p.Lys55SerfsX5. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.164_165delAA variant is not observed in large population cohorts (Lek et al., 2016). Therefore, although this pathogenic variant has not been previously reported to our knowledge, we consider this variant pathogenic.
