This variant lies in the ZNF711 gene (transcript NM_001330574.2) at coding-DNA position 304, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 102 with lysine — a missense variant. Submitter rationale: The E102K variant in the ZNF711 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E102K variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The E102K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret E102K as a variant of uncertain significance.
