Uncertain significance — the classification assigned by GeneDx to NM_004187.5(KDM5C):c.145C>T (p.Pro49Ser), citing GeneDx Variant Classification (06012015): The P49S variant in the KDM5C gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The P49S variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). The P49S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret P49S as a variant of uncertain significance.
