Uncertain significance — the classification assigned by GeneDx to NM_014476.6(PDLIM3):c.1007A>G (p.Glu336Gly), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the PDLIM3 gene. The E336G variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is observed in 7/126720 (0.05%) alleles from individuals of European (Non-Finnish) ancestry in large population cohorts (Lek et al., 2016). This substitution occurs at a position where amino acids with similar properties to glutamic acid (E) are tolerated across species and where glycine (G) is present as the wild type in at least one species. The E336G variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. Finally, in silico analysis predicts this variant is probably damaging to the protein structure/function.
