Uncertain significance — the classification assigned by GeneDx to NM_001370100.5(ZMYND11):c.1259C>A (p.Ser420Tyr), citing GeneDx Variant Classification (06012015). This variant lies in the ZMYND11 gene (transcript NM_001370100.5) at coding-DNA position 1259, where C is replaced by A; at the protein level this means replaces serine at residue 420 with tyrosine — a missense variant. Submitter rationale: The S420Y variant in the ZMYND11 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S420Y variant is not observed in large population cohorts (Lek et al., 2016). The S420Y variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret S420Y as a variant of uncertain significance.
