This variant lies in the STRADA gene (transcript NM_001003787.4) at coding-DNA position 412, where A is replaced by G; at the protein level this means replaces isoleucine at residue 138 with valine — a missense variant. Submitter rationale: The I138V variant in the STRADA gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The I138V variant is observed in 17/33582 (0.05%) alleles from individuals of Latino background in the ExAC dataset (Lek et al., 2016). The I138V variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to Isoleucine are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret I138V as a variant of uncertain significance.
