NM_006904.7(PRKDC):c.1114-1G>A was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the PRKDC gene (transcript NM_006904.7) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1114, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1114-1G>A variant in the PRKDC gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This splice site variant destroys the canonical splice acceptor site in intron 11. It is predicted to cause abnormal gene splicing resulting in an in-frame protein product with an abnormal message. However, in the absence of RNA/functional studies, the actual effect of c.1114-1G>A in this individual is unknown. The c.1114-1G>A variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.1114-1G>A as a variant of uncertain significance.
