Uncertain significance — the classification assigned by GeneDx to NM_006612.6(KIF1C):c.488G>A (p.Arg163Gln), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the KIF1C gene. The R163Q variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The R163Q variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). This substitution occurs at a position where amino acids with similar properties to Arginine are tolerated across species. However, the R163Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. Therefore, based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant.
