Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000302.4(PLOD1):c.1667A>G (p.Tyr556Cys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: PLOD1 c.1667A>G (p.Tyr556Cys) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 7.6e-05 in 251418 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in PLOD1 causing Ehlers-Danlos Syndrome Type VI (7.6e-05 vs 0.0016), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1667A>G in individuals affected with Ehlers-Danlos Syndrome Type VI and no experimental evidence demonstrating its impact on protein function have been reported.
