Pathogenic — the classification assigned by GeneDx to NM_001371928.1(AHDC1):c.2881del (p.His961fs), citing GeneDx Variant Classification (06012015): The c.2881delC variant in the AHDC1 gene causes a frameshift starting with codon Histidine 961, changes this amino acid to a Threonine residue, and creates a premature Stop codon at position 181 of the new reading frame, denoted p.His961ThrfsX181. This variant is predicted to cause loss of normal protein function through protein truncation, as the last 643 amino acids are replaced with 180 incorrect amino acids. Furthermore, the c.2881delC variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Although this pathogenic variant has not been previously reported to our knowledge, its presence is consistent with the diagnosis of Xia-Gibbs syndrome in this individual.
