NM_004999.4(MYO6):c.344_345del (p.Tyr115fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the MYO6 gene (transcript NM_004999.4) at coding-DNA position 344 through coding-DNA position 345, deleting 2 bases; at the protein level this means shifts the reading frame starting at tyrosine residue 115, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.344_345delAT variant in the MYO6 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.344_345delAT variant causes a frameshift starting with codon Tyrosine 115, changes this amino acid to a Serine residue, and creates a premature Stop codon at position 17 of the new reading frame, denoted p.Tyr115SerfsX17. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.344_345delAT variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.344_345delAT as a pathogenic variant.
