NM_001142864.4(PIEZO1):c.6079G>A (p.Val2027Met) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the PIEZO1 gene (transcript NM_001142864.4) at coding-DNA position 6079, where G is replaced by A; at the protein level this means replaces valine at residue 2027 with methionine — a missense variant. Submitter rationale: The V2027M variant in the PIEZO1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The V2027M variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The V2027M variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret V2027M as a variant of uncertain significance.
