Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_030777.4(SLC2A10):c.212G>T (p.Cys71Phe), citing Ambry Variant Classification Scheme 2023: The p.C71F variant (also known as c.212G>T), located in coding exon 2 of the SLC2A10 gene, results from a G to T substitution at nucleotide position 212. The cysteine at codon 71 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
