Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.2429T>A (p.Met810Lys), citing Ambry Variant Classification Scheme 2023: The p.M810K variant (also known as c.2429T>A), located in coding exon 14 of the MYLK gene, results from a T to A substitution at nucleotide position 2429. The methionine at codon 810 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species; however, lysine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
