Uncertain significance — the classification assigned by GeneDx to NM_000041.4(APOE):c.148C>A (p.Arg50Ser), citing GeneDx Variant Classification (06012015): The R50S variant in the APOE gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R50S variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R50S variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret R50S as a variant of uncertain significance.
