This variant lies in the DARS2 gene (transcript NM_018122.5) at coding-DNA position 188, where G is replaced by A; at the protein level this means replaces glycine at residue 63 with aspartic acid — a missense variant. Submitter rationale: The G63D variant in the DARS2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The G63D variant is observed in 40/10406 (0.38%) alleles from individuals of African background in the ExAC dataset and no individuals were reported to be homozygous (Lek et al., 2016). The G63D variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret G63D as a variant of uncertain significance.
